Modifications in vapor diffusion resistence of leaves and water relations in barbados cherry plants under water stress Rev. Bras. Fisiol. Veg.
NOGUEIRA,REJANE J. MANSUR C.; MORAES,JOSÉ ANTÔNIO P. V. DE; BURITY,HÉLIO ALMEIDA; BEZERRA NETO,EGÍDIO.
Young sexually and assexually propagated Barbados cherry plants were submitted to water deficit (20 days without irrigation). During this period the accumulation of proline, water potential of branches, osmotic potential, the relative water content of leaves, the leaf diffusive resistance, the transpiration rate and leaf temperature in the cuvette were determined. In addition, photosynthetically active radiation (PAR) and vapor pressure deficit (VPD) were measured in the porometer cuvette. The concentration of proline for both types of plants began to increase on the fifth day without watering, and reached 38.1 times the concentration in the control plants grown from seeds and 26.4 times the concentration in grafted plants on the tenth day without...

An improved method for transformation of lettuce by Agrobacterium tumefaciens with a gene that confers freezing resistance BABT
Pileggi,Marcos; Pereira,Albanin Aparecida Mielniczki; Silva,Joandrei dos Santos; Pileggi,Sônia Alvim Veiga; Verma,Desh Pal S..
An efficient method for constructing transgenic lettuce cultivars by Agrobacterium tumefaciens was described by Torres et al., 1993. In the present work, an improvement of the above procedure is described and applied to transform the cultivar Grand Rapids with a mutated P5CS gene. The major modifications were concerned with turning more practical the transformation and regeneration protocols. Also we tried to improve transformation steps by increasing injured area in explants and prolonging co-cultivation with Agrobacteria (in larger concentration). A more significant selective pressure was used against non-transformed plants and bacteria. In these work we were concerned to obtain T1 and T2 seeds. The P5CS gene codes for a delta¹-pyrroline-5-carboxylate...

Proline content and protein patterns in Eucalyptus grandis shoots submitted to high and low temperature shocks BABT
Souza,Gustavo M.; Cardoso,Victor J. M.; Gonçalves,Antonio N..
Proline content and protein patterns changes in response to temperature shocks of both acclimated and non acclimated E. grandis shoots cultivated in vitro were investigated. Analysis of soluble proteins through SDS-PAGE and proline were carried out after 12h at 12ºC (cold acclimation) or 33ºC (heat acclimation), and immediately after temperature shocks at 41ºC and 0ºC. Analyses were also performed 24h after the temperature shocks (recovery period). Temperature treatment at 0ºC did not change soluble protein patterns both in acclimatized and non acclimatized plants, whereas cold temperature induced high proline levels, which kept relatively high after recovery period. Three novel, probably HSPs, proteins (90.5, 75 and 39 kDa) were observed in both...

The effects of salt stress on growth, nitrate reduction and proline and glycinebetaine accumulation in Prosopis alba Braz. J. Plant Physiol.
Meloni,Diego Ariel; Gulotta,Marta Rosalía; Martínez,Carlos Alberto; Oliva,Marco Antonio.
Prosopis alba (algarrobo) is one of the most important salt-tolerant legumes used in the food and furniture industries. The effects of salinity on some growth and physiological parameters in algarrrobo seedlings were investigated. 17-Day-old seedlings were subjected to three salt treatments by adding NaCl to the growth medium in 50 mmol.L-1 increments every 24 h until the final concentrations of 0, 300 and 600 mmol.L-1 were reached. Only the highest NaCl concentration affected all of the considered parameters. Thus, 600 mmol.L-1 NaCl caused a significant reduction in root and shoot growth, but an increase in the root/shoot ratio. Leaf relative water content, nitrate content and nitrate reductase activity in leaves and roots were also decreased. At 300 and...

Effects of substrate water potential in root growth of Agave salmiana Otto ex Salm-Dyck seedlings Biol. Res.
PEÑA-VALDIVIA,CECILIA B; SÁNCHEZ-URDANETA,ADRIANA B.
The objective of this study was to test the hypothesis that root of maguey (Agave salmiana Otto ex Salm-Dyck) seedlings reacts during the first 24 h to low substrate water potential (Yw), by anatomical modifications. Three-4 cm root length seedlings were planted in vermiculite for 24 h at Yw between -0.03 and -2.35 MPa. Root dimensions, proline content and anatomy were evaluated. Substrate ψw between -0.65 and -2.35 MPa did not significantly affect longitudinal root growth. However, proline content significantly increased from 1.6 to 2.1 emoles mg-1. Significant reductions of transverse root area (41 %), thickness of mucilage covering the epidermis (47 %), thickness of epidermis (between 15 and 46 %), area of the parenchyma (between 35 and 41 %)...

Study of potential impacts of using sewage sludge in the amendment of desert reclaimed soil on wheat and jews mallow plants BABT
Mazen,Ahmed; Faheed,Fayza A; Ahmed,Atef F.
This investigation was conducted to study the impacts of using sewage sludge at different concentrations (0, 10, 25, 50, 75%) in amendment of desert reclaimed soil properties and some physiological aspects in wheat and jews mallow plants. Generally adding sewage sludge to desert soil improved the soil texture, raised the organic matter contents, water holding capacity and lowered pH value. The contents of NPK gradually increased as the ratio of sewage sludge increased. The fresh and dry weights and biosyntheses of pigment contents of the variously treated test plants were increased by increasing the sewage sludge levels in the soil. Also, total carbohydrate and protein contents of sewage sludge-treated test plants were positively affected. With respect to...

Glomus intraradices improved salt tolerance in Prosopis alba seedlings by improving water use efficiency and shoot water content Braz. J. Plant Physiol.
Scambato,Agustina A.; Echeverria,Mariela; Sansberro,Pedro; Ruiz,Oscar A.; Menéndez,Ana Bernardina.
The present work was aimed at testing the hypothesis that mycorrhizal Prosopis alba, an economically important tree species worldwide, presents increased salt-tolerance compared with non-mycorrhizal ones and at gaining insight into the possible mechanisms underlying that improvement. For this purpose, a randomized complete block experiment with two factors: mycorrhizal treatments with or without the arbuscular fungus Glomus intraradices and two salinity levels, 0 and 200 mM NaCl was performed. Plant growth in P. alba plants colonized by G. intraradices was less affected by salinity than that in non-arbuscular mycorrhizal (AM) plants, indicating that mycorrhizal colonization turned P. alba more tolerant to salinity. Photosynthesis was reduced by salinity in...
